Carell, youngest of four brothers, was born in Acton, Massachusetts, the son of Harriet T. (née Koch), a psychiatric nurse, and Edwin A. Carell, an electrical engineer.[2][3][4] Carell's maternal uncle was Stanley Koch, a glassblower who worked with Allen B. DuMont to create cathode ray tubes.[5] Carell's paternal grandfather was Italian;[2] his father was born with the surname "Caroselli", later shortening it to "Carell".[4] Carell was educated at The Fenn School and Middlesex School, where he graduated in 1980, and took part on the Junior varsity Dance team [dancer, number 22] and lacrosse teams in Concord, as well at Denison University in Granville, Ohio.

In 1996, Carell was a cast member of the briefly aired The Dana Carvey Show. Along with fellow cast member Stephen Colbert, Carell provided the voice of Gary, half of "The Ambiguously Gay Duo", the Robert Smigel-produced animated short.

Andy Stitzer (Steve Carell) is the eponymous 40-year-old virgin. While being a very well-meaning, highly neurotic and something of a stereotypical nerd, he lives alone, collects action figures, enjoys video games, framed a poster of eighties rock band Asia, and his social life seems to consist of watching Survivor with his elderly neighbors.
Andy works in the stockroom at an electronics store called SmartTech. His co-workers include the affable David (Paul Rudd), who is tormented by memories of his old girlfriend Amy (Mindy Kaling); the burly Cal (Seth Rogen), a crude, self-proclaimed novelist and stoner; and Jay (Romany Malco), a self-styled ladies' man. When a friend drops out of a poker game, they reluctantly invite the withdrawn Andy to join them. Andy turns out to be surprisingly good at poker due to his playing the game online, but when the conversations turns to past sexual exploits, they quickly realize Andy is still a virgin, and resolve to help him lose his virginity.

Newly elected to Congress, former local TV newsman Evan Baxter (Steve Carell) leaves Buffalo behind and shepherds his family to suburban northern Virginia. His congressional campaign declared that he would change the world, but he never stated how. Once he reaches his new home and job, strange things start to happen. Animals start following Evan around. He grows a beard that won't go away no matter how many times he shaves. Eight vacant lots in Evan's neighborhood are purchased in Evan's name. Tools used in Biblical times, and wood planks, are sent to his house. And the number 614 appears everywhere he goes. Evan soon learns that the number is a verse in the Bible. In the book of Genesis, 6:14 is the verse in which God instructs Noah to build the ark. Soon after, God (Morgan Freeman) appears and mysteriously commands him to build an ark because a flood is coming. His family comes to believe he is having an extraordinary mid-life crisis, but soon thinks he is truly onto something of Biblical proportions. Although they believe it's a bit crazy, Evan's boys help him build the ark. His wife Joan (Lauren Graham), doesn't want to help. She feels it's too embarrassing. God tells Evan the flood will come September 22 mid-day.

Dan Burns (Steve Carell), is a newspaper columnist and a widowed father of three girls: Jane, Cara, and Lily. Jane has a driver's license, but Dan does not allow her to drive his car. Cara is very much in love with her boyfriend, but Dan thinks she is too young for that and keeps them apart. Cara is furious and repeatedly calls her father a murderer of love. Dan and his daughters take a trip to the home of his parents and visits his family, including his brother Mitch (Dane Cook). The morning after his arrival, Dan is encouraged by his mother to go into town for a bit to give his daughters some space. Dan visits a bookstore and is mistaken for an employee by a beautiful woman, Marie (Juliette Binoche). Dan and Marie strike up a conversation and continue their talk over coffee. They lose track of time, but both are clearly attracted to the other, although, as Marie tells Dan, she has a boyfriend. Dan returns to his parents and is surprised when Mitch introduces his girlfriend: Marie.

Bernie Kopell, who played Siegfried in the original series, makes a cameo.
